I think the best definition, purpose  and example are itself given in PMBOK P197 section 8.1.2.5.

I beleive, for understanding and exam point of view, both, it is the best one.

Which line you cant get from that, let me know.

--------------------------------------------

In my words,  I explain here:

Design of Experiments: To design a framework, (like any structural design or computer programming), for a series of experiments (number of experiments)

or

A designed framework for conducting number of expeiments is called DOE.

--------------

What is purpose:  To establish (invent) the best combination from different factors (or variables), of an subjected object, to obtain the desired degree of requirement or result.

-------------------

For example: example of pmbok :

To obtain desired degree of comfort (no jerk or thrust) for passangers in a car cab, A series of experiments is designed. They do the experiments changing , various factors of car, like Tire pressure and shock- up spring, strength, and at the end they establish a desired combination (optimized) with a certain degree of relief. So this is a DOE.

Hope it is clear -- now read PMBOK FILL THE GAPS.
